  Professor Donald L. Gill is a distinguished Professor and Chair of Cellular and Molecular Physiology,from the Pennsylvania State University. His research group centers on the essential mediator of calcium signal, calcium release-activated calcium (CRAC) channel, and have published more than 50 papers, including milestone discoveries in the last 10 years. This talk will focus on the physiological role of calcium signal mediated by CRAC channels, and the STIM-Orai coupling interface, which will shed light on the gating mechanisms of CRAC channels.

  报告摘要:

  Orai1-mediated “store-operated” Ca2+ signals control gene expression, growth, secretion, and motile responses universally among cells and are implicated in a spectrum of immunological, muscular and inflammatory disease states. Despite intense study, the molecular nature of the STIM-Orai1 coupling interface and the mechanism of Orai1 channel gating remain obscure. We have identified the Orai-activating sites on STIM1 by comparing the distinct Orai-coupling domains in STIM1 and STIM2, and discovered that STIM1 dimers undergo unimolecular coupling to activate Orai1 channels. This talk will focus on the physiological role of calcium signal mediated by CRAC channels, and the STIM-Orai coupling interface, which will shed light on the novel gating mechanism of CRAC channels.
